Abstract

Burcucumber (Sicyos angluatus) is a representative ecosystem-disturbing plant in Korea and currently widely spread throughout the country. A sclerotia-forming fungus with moderate host selectivity, Sclerotinia trifoliorum BWC98-105, was tested in the laboratory, green house and natural habitat for its pathogenicity to burcucumber. When mycelial culture fragment was inoculated to burcucumber seedlings under the green house condition, mycelial growth was observed in the following day, and then resulted in the onset of wilting from 5 days after inoculation (DAI). Its characteristic sclerotia as a sign was observed from 7 DAI, and thus plants turned into dark-brown color at the bottom of stem of burcucumber that was eventually blighted at 14 DAI. Similar visible symptoms were observed in natural habitat. Based on the results of showing typical blight symptom to burcucumber and the sign of sclerotia, we report S. trifoliorum BWC98-105 causing stem blight against burcucumber. Its globular pellet was considered of having quite potential as a bioherbicide to control burcucumber in Korea.

가시박(Sicyos angulatus)은 북아메리카 원산의 1년생 박과식물로 1994년에 최초로 보고되었으며, 환경부에서 지정한 14개 외래생태계교란식물 중 대표적인 식물로 특히 수변지역의 식물생태계를 심각하게 교란시키며 전국적으로 확산되어 있다(Kang, 2014; Moon 등, 2007). 가시박의 생태계교란은 빠른 생육과 덩굴손으로 공존하는 식물의 광합성을 억제하고 억센 가시에 의한 소동물의 이동성을 제한하여 발생지역의 식물상을 단순화시킴으로써 이루어진다(Kang, 2014). 현재 가시박은 발생지역의 경운 또는 식물체의 제거와 같은 물리적인 방법으로 일부만 관리되는 실정이고, 천연물을 사용한 화학적인 방법이나 미생물을 이용한 생물학적인 방법은 아직 연구단계에 머무르고 있다.

국내에서 가시박에 대한 병원성을 보이는 미생물은 매우 드물다. Kang (2014)은 Cercospora citrullina, Pseudocercospora cantuariensisStagonosporopsis cucurbitacearum 3종을 보고하였고 이들을 생물학적 방제용 미생물로 검토하였다. 그러나 이들 진균은 배양이 어렵고 생존 기간이 짧아 실제 적용하기에는 어려운 점이 있다. 국내에서 기주선택성이 있으면서 균핵(sclerotia)을 형성하는 특징을 가진 진균으로 자낭균류인 Sclerotinia trifoliorum BWC98-105 균주가 잔디밭의 클로버 방제용 바이오제초제로서 보고된 바 있다(Hong 등, 2002, 2004). 이 균주는 주로 콩과식물과 일부 잡초성 식물에서 병원성이 있고, 벼, 보리, 옥수수, 밀, 잔디 등의 작물에는 병원성이 없었다. 해외에서도 분류학적으로 유사한 S. minorS. sclerotiorum을 이용해서 바이오제초제로서 사용하거나 개발하고 있다. S. minor IMI344141 균주는 잔디밭에 발생하는 광엽 잡초를 관리하기 위한 바이오제초제로 캐나다에서 등록되었다(Pest Management Regulatory Agency, 2010). S. sclerotiorum은 엉겅퀴류의 잡초를 효과적으로 제거할 수 있다고 보고되었다(Skipp 등, 2013).

가시박의 생물학적 방제는 환경친화적인 관리방법이 될 수 있기 때문에 연구개발의 필요성이 높다. 또한 균핵은 생육에 불리한 환경조건에서도 생존할 수 있으므로 바이오제초제로 개발될 경우, 생산∙저장 및 유통과정에서 편리한 장점을 가지고 있다. 따라서 본 연구에서는 균핵형성균 S. trifoliorum BWC98-105 균주의 가시박에 대한 병원성을 확인하고, 가시박의 생물학적 관리를 위한 바이오제초제로서의 가능성을 타진하고자 하였다.

균사 배양체는 S. trifoliorum BWC98-105를 감자한천배지(potato dextrose agar, Difco, Detroit, MI, USA)에서 약 1주일 동안 25°C에서 배양하여 균핵이 형성되기 이전에 사용하였다. 균핵은 상기 배양조건에서 약 4주일 동안 배양하여 배지 위에 형성된 균핵을 수확하고 건조하여 사용하였다(Fig. 1A). 또한 구형 펠렛은 일반적인 종자 코팅 방법을 응용하여, 코팅기에 균핵과 증량제 등 성분을 넣고 회전시키면서 살수하여 제조하였다(Kim 등, 2013). 각 구형 펠렛에는 한 개의 균핵이 포함되도록 제조하였다(Fig. 1B). 실내 또는 온실 포트 실험을 위한 식물체로는 2017년 11월 경기도 성남지역에서 채종된 가시박 종자를 키워 사용하였다. 가시박 종자는 휴면성이 높기 때문에, 종피를 벗긴 후 페트리 디쉬의 여과지 위에 놓고 지베렐린(90% Gibberellin A3, Sigma-Aldrich, St. Louis, MO, USA) 100 μg/ml 수용액을 매일 분사해 휴면타파하고 발아시켰다. 이 방법을 사용하면 대부분의 종자가 3–5일 사이에 발아되었다. 주근이 3cm 정도 자란 후 균사 배양체 조각을 근접하게 위치시킨 후 뚜껑을 덮고 상온에 보관하였다. 발아된 가시박을 포트(직경 8.5 cm, 높이 6.5 cm)에 식재한 후 2–3 본엽 단계까지 온실에서 키웠다. 감자한천배지에서 1주일 동안 25°C에서 배양된 균사 배양체 조각(1 cm×1 cm×0.5 cm)을 가시박 줄기의 지저부에 근접하게 위치시켜 접종하였다. 온실실험에서 시험군마다 10개의 포트를 사용하였다. 접종원은 균사 배양체 조각과 구형 펠렛(직경 ∼4 mm)을 사용하였다. 균사 배양체 조각이나 구형 펠렛은 가시박 유묘의 지저부에 근접하게 위치시켰다. 가시박은 각 포트에서 저면관수하며 키웠고, 경엽부위를 원통형 비닐(직경 12 cm)로 덮어 다습한 조건을 제공하였다. 현장실험은 2018년 경기도 성남지역에서 자연적으로 형성된 가시박 군락에서 실시하였다. 가시박은 4월 초부터 본격적으로 출아되기 시작했고, 1차 살포 시점에서 가시박 줄기는 1 m를 넘으면서 본격적으로 지표면 위에서 덩굴로 포복하기 시작하였다. 가시박의 발생밀도가 높은 지역을 선정하여 시험구(5 m×4 m)를 구획하였다. 구형 펠렛은 약 한달 간격으로 3회(1차 5월 20일, 2차 6월 30일, 3차 7월 15일)에 걸쳐 살포했고, 살포량은 10 g/m2 수준이었고, 3회(1차 6월 24일, 2차 7월 29일, 3차 8월 26일)에 걸쳐 가시박의 피복도와 S. trifoliorum BWC98-105의 병원성을 조사하였다. 피복도는 시험구에서 무처리 대조군의 면적대비 가시박의 표면적 비율로 산정하였다.

실내실험에서 균핵형성균 S. trifoliorum BWC98-105의 균사생장은 균사 배양체의 접종 후 익일부터 육안으로도 관찰되었다. 접종 후 5일 시점에서 가시박 유묘가 시들기 시작했다. 접종 후 7일부터 특징적인 표징(sign)인 균핵의 형성이 관찰되었고, 14일 시점에서는 식물체가 고사되고 대부분의 균핵이 성숙되고 진한 갈색으로 변했다. 이로부터 재분리된 균핵의 배양적 특성을 기준으로 동일한 균핵형성균임을 확인하였다. 따라서 S. trifoliorum BWC98-105는 가시박에 대한 병원성이 있다고 추정하였다. 온실실험에서도 균사 배양체의 접종 후 3일부터 가시박의 줄기에서 자라는 균사를 관찰할 수 있었다(Fig. 2A). 접종한 10개체 중 6–9개체에서 균사생장이 관찰되었다. 접종 후 초기에 가시박 줄기에 정착하지 못한 전염원은 이후에도 병이 발생하지 않았다. 접종 후 14일 시점에서 가시박의 줄기에서 균핵이 형성되기 시작했고 위조현상이 관찰되었다(Fig. 2B). 접종 후 21일 시점에서는 갈색의 균핵이 관찰되었고 가시박은 고사되었다. 이와 같이 S. trifoliorum BWC98-105는 실내실험과 마찬가지로 온실실험에서도 가시박 유묘에 대한 병원성을 보였다. 또한, 균사 배양체뿐만 아니라 구형 펠렛을 처리한 경우에도 병원성이 확인되었다. 현장실험에서 가시박에 대한 병원성은 2018년 7월 15일(1차 살포 후 62일, 2차 살포 후 15일)에 처음으로 관찰되었다. Table 1에 제시된 바와 같이 1차 조사시점(6월 24일)에서는 피복도를 기준으로 차이를 관찰할 수 없었다. 1차 조사 시점에서 무처리 대조군과 구형 펠렛 시험군에서 피복도의 차이가 관찰되지 않았다. 이는 가시박 줄기가 1–3 m 수준으로 자랐지만 지표면을 덮을 만큼 충분히 자라지 않았고, 평균기온이 21.3°C로 장마 시작 이전에 온도와 습도가 낮았기 때문이라고 추정되었다. 2차 조사시점(7월 29일)부터는 시험군의 피복도가 무처리 대조군보다 낮았다. 또한 가시박의 위조 또는 고사현상이 본격적으로 관찰되었다(Fig. 3). 또한 지표면을 포복하는 가시박 줄기에서 균사 생장, 줄기마름 증상 및 균핵 형성이 관찰되었다. 3차 조사시점(8월 26일)에서도 시험군에서 가시박의 피복도는 낮게 유지되었다. 2차 조사시점 이후부터 약효가 관찰된 이유는 가시박의 추가적인 줄기생장과 장마철 고온 다습한 조건으로 발병이 유리한 환경이 조성되었고, 추가 살포에 의해 균핵형성균의 밀도가 높아졌기 때문이라고 사료되었다. 균핵형성균에 의해 가시박이 고사되었음에도 불구하고 동일한 공간에서 환삼덩굴, 명아주, 까마중, 강아지풀 등 비표적 식물이 정상적으로 자라고 있었는데, 이를 통해 균핵형성균의 기주선택성을 일정 수준 확인할 수 있었다.

이와 같이 균핵형성균 S. trifoliorum BWC98-105는 실내, 온실 및 현장실험에서 모두 가시박에 대한 병원성을 나타냈다. 따라서 S. trifoliorum BWC98-105의 가시박에 대한 줄기마름병(stem blight) 증상을 보고하고자 한다. 또한 균핵을 포함하는 구형 펠렛은 균핵의 저장 안정성과 펠렛의 사용 편리성을 고려할 때 바이오제초제로서 개발될 가능성이 높다고 판단되었다. 또한 국내에서 자생하는 토착미생물을 이용한 생물학적 방제는 약효가 오랫동안 지속될 수 있고, 자연생태계의 조절 능력에 의해 생물학적인 균형을 유지할 수 있는 장점이 있다고 볼 수 있다. 향후 가시박과 같은 생태계 교란식물에 대한 제초효과를 체계적으로 확립하고, 기주선택성에 의한 주변 식물생태계에 대한 영향을 정립할 필요가 있다.

요 약

가시박(Sicyos angulatus)은 대표적인 외래 생태계교란식물로 전국적으로 확산되어 발생하고 있다. 국내에서 기주선택성이 있으면서 균핵을 형성하는 Sclerotinia trifoliorum BWC98-105 균주의 균사 배양체와 구형 펠렛으로 가시박에 대한 병원성을 실내, 온실 및 현장실험에서 확인하였다. 온실실험에서 균사 배양체는 접종 후 익일부터 균사생장이 관찰되었고, 5일부터 시들기 시작하였고, 7일부터 특징적인 표징(sign)인 균핵의 형성이 관찰되었고, 14일 시점에서는 식물체가 고사되고 대부분의 균핵이 성숙한 갈색으로 변했다. 이러한 현상은 현장실험에서도 확인되었다. 따라서 S. trifoliorum BWC98-105의 가시박에 대한 줄기마름병(stem blight) 증상을 보고하고자 한다. 또한 균핵형성균 S. trifoliorum BWC98-105의 균핵으로 제조된 구형 펠렛은 향후 가시박 관리를 위한 바이오제초제로써 가능성이 높다고 사료되었다.
