Abstract

Recently, global warming and drastic climate change are the greatest threat to the world. The climate change can affect plant productivity by reducing plant adaptation to diverse environments including frequent high temperature; worsen drought condition and increased pathogen transmission and infection. Plants have to survive in this condition with a variety of biotic (pathogen/pest attack) and abiotic stress (salt, high/low temperature, drought). Plants can interact with beneficial microbes including plant growth-promoting rhizobacteria, which help plant mitigate biotic and abiotic stress. This overview presents that rhizobacteria plays an important role in induced systemic resistance (ISR) to biotic stress or induced systemic tolerance (IST) to abiotic stress condition; bacterial determinants related to ISR and/or IST. In addition, we describe effects of rhizobacteria on defense/tolerance related signal pathway in plants. We also review recent information including plant resistance or tolerance against multiple stresses (bioticabiotic). We desire that this review contribute to expand understanding and knowledge on the microbial application in a constantly varying agroecosystem, and suggest beneficial microbes as one of alternative environment-friendly application to alleviate multiple stresses.

서론

지구 온난화를 비롯한 기후변화는 1950년대 이후 지속적으로 이어지고 있으며, 이는 전 지구 평균 온도 상승, 고온 또는 저온의 극한 기온 변화, 이상 건조 또는 호우 발생을 가져왔다(Hansen 등, 2012; Stocker 등, 2013). Intergovernmental Panel on Climate Change 5차 평가 보고서의 시나리오에 따르면 이산화탄소 누적에 따른 지구 온난화 영향으로 다수의 지역에서 폭염의 발생 빈도와 지속기간이 증가하고 있으며, 강수 발생빈도와 강도 역시 증가하는 추세이다. 이러한 폭염, 가뭄, 홍수와 같은 극한 기후현상으로 일부 생태계, 특히 농업생태계는 심각하게 영향을 받고 있으며, 식물의 생산성에 상당히 부정적인 영향을 주는 것으로 알려져 있다(Boyer, 1982; Bray 등, 2000). 특히, 식물은 생식기간 중에 고온에 노출되면 생산량이 크게 감소하며(Forni 등, 2016; Verslues 등, 2006), 빈번한 고온 및 다습 조건은 다양한 작물에 피해를 야기하고 있다(Kal 등, 2015; Kang 등, 2016b). 이러한 이상기후로 인해 열대 및 온대지역에서 밀, 쌀 및 옥수수의 수확량이 감소할 것으로 추정되며, 멸종 가능성이 있는 식물 수가 증가하는 등 식량안보에 위협이 될 가능성이 있다(Field 등, 2014).

식물은 재배기간 중에 다양한 환경변화뿐만 아니라 병원균의 공격과 같은 생물 스트레스에 노출되어 있다. 식물이 겪는 생물 스트레스 중 세균, 진균, 바이러스 등의 다양한 병원성 미생물에 의해 발생하는 것은 시간, 장소, 유전자형에 따라 밀도나 특성에 영향을 받을 수 있기 때문에 이를 방제하는 데 어려움이 있다(Strange와 Scott, 2005). 이러한 병원균의 공격으로부터 식물을 보호하기 위하여 농약을 사용하는 화학적 방제방법, 경종 · 물리적 방법, 식물 뿌리 주위에 생존하는 미생물 중 식물 생장 촉진에 관여하는 식물생장촉진근권세균(plant growth-promoting rhizobacteria, PGPR) 등의 유용한 근권미생물을 이용한 생물학적 방제 방법 등이 연구되고 있으나(Bakker와 Schippers, 1987; Kloepper 등, 2004), 주로 개별 스트레스 요인과 영향을 감소시키는 방법으로 연구되어 왔다(Ramegowda와 Senthil-Kumar, 2015). 그러나 실제 식물은 극한 온도변화, 과도한 건조 또는 과습, 염류 집적 토양 등 환경 요인과 병원균, 해충, 초식동물 등의 공격에 의한 생물 요인이 동시에 일어나는 복합적인 환경에서 재배되는 것이 대부분이며(Atkinson과 Urwin, 2012), 앞으로는 복합 스트레스에 의한 피해가 더 증가할 것으로 예상되고 있다(Kissoudis 등, 2014; Suzuki 등, 2014). 환경 요인은 병원균과 같은 생물 요인에도 영향을 미치는데, 고온 조건에서 일부 고온성 병원균의 확산이 더 용이해지며 이처럼 병원균이나 해충의 생육조건이 기후변화로 충족될 경우 그 피해는 더 심각할 수 있다(Atkinson과 Urwin, 2012; Bale 등, 2002; Goel 등, 2008; Madgwick 등, 2011). 또한 다양한 환경 스트레스에 의해 식물이 병원균에 대해 갖고 있는 기본적인 방어 능력이 약화되거나 병원균에 대한 감수성이 변화하여 병이 더 많이 발생하기도 한다(Atkinson과 Urwin, 2012; Goel 등, 2008). 실제 최근 국내 배추 생산은 재배 초기 고온과 가뭄, 수확기 잦은 강우로 인한 다습조건으로 무름병에 의한 피해가 심각했으며, 이는 곧 배추의 생산량 감소와 시장가격 상승으로 이어졌다(Lee 등, 2016).

이처럼 생물 · 환경적 스트레스의 복합적인 피해 사례는 빈번하게 발생하고 있으며, 이러한 피해에 대비하기 위해 복합 스트레스에 대한 내성을 갖는 식물을 육종하기 위한 기작 연구가 진행되고 있다(Atkinson 등, 2013; Bostock 등, 2014; Kissoudis 등, 2014; Rasmussen 등, 2013). 또한 화학 농약 등을 이용한 기존의 방제 방법의 대안으로 환경적으로 더 안전하고 생태계의 균형을 깨트리지 않는 미생물을 이용한 식물 면역 활성화 등에 대한 연구가 부각되고 있다(Maksimov 등, 2015). PGPR을 포함한 다양한 근권 유용미생물은 병원균의 침입에 대해 식물에 저항성을 유도하는 유도전신저항성(induced systemic resistance, ISR)을 일으키며(Okmen과 Doehlemann, 2014; Wei 등, 1991), 환경 스트레스에 대항하기 위해 유도전신내성(induced systemic tolerance, IST)을 이용하여 식물에 내성을 유도하기도 한다(Yang 등, 2009). 병원성 미생물에 의해 유도되는 전신획득저항성(systemic acquired resistance, SAR) 역시 식물의 대표적인 유도 저항성 반응으로 알려져 있다(Gozzo와 Faoro, 2013). SAR은 병원균에 의해 광범위하게 유도될 수는 있지만, 병원균의 괴사반응에 의해 저항성 반응이 유도되기 때문에 어떤 외적 증상 없이 일어나는 ISR과 비교했을 때 차이가 있다(Fu와 Dong, 2013; van Loon 등, 1998). 또한 ISR은 SAR에 비해 식물에 약해와 같은 증상 없이 저항성 반응을 유도한다는 장점이 있다(Lee 등, 2015). Heil (1999)의 연구에서는 식물은 가지고 있는 한정된 에너지에서 저항성 유도에 과도한 에너지를 사용할 경우 생장에 필요한 에너지가 부족하여 생장이 감소하는 식물에너지보존이론(allocation fitness cost)을 제안하였다. PGPR 등의 유용한 근권미생물에 의한 식물의 저항성 유도는 식물의 에너지 효율을 극대화하기 위하여 병원균의 침입 시에만 빠르고 강하게 반응하여 효과를 내는 방어프라이밍(defense priming)을 통해 식물 생존에 도움을 줄 수 있다(Prime-A-Plant Group, 2006). 그러므로, 유용미생물에 의한 방어프라이밍은 저항성 유도에 따른 식물의 한정된 에너지 소비를 최소화하기 때문에, 이를 이용한 복합스트레스에 대한 농업적 적용방안과 기전에 대한 연구가 더 필요할 것이다(Cook 등, 1995; Dimkpa 등, 2009; Pieterse 등, 2014).

본 논문에서는 각각의 또는 복합적인 생물 · 환경적 스트레스에 대응하여 PGPR 등의 유용한 근권미생물에 의한 식물 저항성 또는 내성 유도 역할과 영향을 주는 요인(Table 1), 이에 의한 식물의 신호전달 체계의 변화에 대해 소개하고(Fig. 1), 이를 통해 급격한 환경변화에 의한 생물 · 환경의 복합적인 스트레스에 대한 식물의 피해를 경감시키기 위한 농업적 활용 방안을 모색하고자 하였다.

식물의 생물적 스트레스에 대한 미생물의 역할

식물의 생물적 스트레스 영향 감소

식물의 뿌리 주위에서 서식하며 식물의 생육을 촉진하는 PGPR 등의 근권미생물은 병원균에 의한 생물적 스트레스를 감소시키는 것으로 알려져 있다(Bakker 등, 2007; Kloepper와 Schroth, 1981; Maksimov 등, 2015). PGPR 등의 근권미생물은 병원균과 같은 서식처에서 양분 및 생장에 대해 경쟁을 하거나(Beneduzi 등, 2012), 병원균에 대한 항생효과를 가지는 대사물질들을 합성 및 분비하는 등 직 · 간접적으로 병원균의 생장을 저해시킴으로써 식물병을 억제한다(Maksimov 등, 2015). PGPR 등의 근권미생물에 의하여 Rhizoctonia solaniPythium에 의한 병발생이 감소하였고(Kloepper, 1991) 세균성 병원균의 생장도 감소시켰다(Kloepper, 1983). 반면, PGPR은 식물에 저항성을 유도하여 병원균에 의한 생물적 스트레스를 감소시키기도 하는데, 곡류, 박류, 콩과 및 가지과 식물 등 25가지 이상의 식물에서 ISR을 통해 다양한 병원균에 대하여 식물에 저항성을 유도하였다(van Loon 등, 1998; Wei 등, 1991). 식물은 미생물이 가지는 고유의 분자적 패턴을 인식하여 다양한 경로의 면역반응을 일으키는데(Jones와 Dangl, 2006), 이때 식물이 인식하는 미생물의 패턴을 미생물 관련 분자 패턴(microbe-associated molecular pattern, MAMP), 그 미생물이 병원균일 경우, 병원균 관련 분자 패턴(pathogen-associated molecular pattern, PAMP)이라고 한다(Dodds와 Rathjen, 2010; Newman 등, 2013). 근권미생물이 가지는 분자패턴도 식물에 저항성을 유도하는 요인이 될 수 있다고 알려졌다(Pieterse 등, 2014; van Loon, 2007). PGPR인 Pseudomonas putida WCS358의 flagellin과 P. fluorescens WCS417r의 lipopolysaccharide (LPS)는 대표적인 MAMP로써 애기장대에 ISR을 유도하여 세균성병원균 P. syringae pv. tomato DC3000에 대한 저항성을 증가시키는 것으로 보고되었다(Meziane 등, 2005). 최근 연구에서는 근권미생물의 MAMP에 의해 콩의 Xanthomonas axonopodes pv. glycines에 대한 저항성 증가와 함께 2차 대사산물 isoflavone의 생성이 함께 증가하는 것이 보고되었다(Algar 등, 2014). Niu 등(2016)의 연구에 따르면 Bacillus cereus AR156는 P. syringae pv. tomato DC3000에 대한 애기장대의 SAR을 유도하며, flagellin을 인식하는 수용체 FLG22-INDUCED RECEPTOR-LIKE KINASE 1 (FRK1)의 발현량을 증가시켜 P. syringae pv. tomato DC3000에 대한 저항성을 유도한다고 보고하였다.

생물적 스트레스 감소에 영향을 주는 미생물 결정 인자

① 항생물질(antibiotics)

항생물질은 세균이 만들어내는 비교적 작은 분자량의 유기 화합물로 미생물의 생육이나 발달을 억제 및 지연시키며, 병원균의 세포벽 합성을 억제시켜 세포막 구조에 영향을 주거나 리보솜 소단위(small subunit of the ribosome)의 개시복합체(initiation complex) 형성을 억제한다(Maksimov 등, 2011). 이와 함께 항생물질은 식물의 유도 저항성 반응 중 하나인 ISR을 유도함으로써 간접적으로 식물의 저항성 반응에 관여하기도 한다(De Vleesschauwer와 Höfte, 2009; Whipps, 2001). 대표적인 항생물질인 2,4-diacetylphloroglucinol (DAPG)은 P. fluorescens CHA0에 의해 생성되어 노균병을 유도하는 Hyaloperonospora arabidopsidis에 대한 애기장대의 저항성을 유도하며, 선충 Meloidogyne javanica에 대한 토마토의 저항성을 증가시킨다고 보고되었다(Iavicoli 등, 2003; Siddiqui와 Shaukat, 2003). P. fluorescens strain가 생성한 DAPG에 의해 애기장대에서 P. syringae pv. tomato DC3000에 대한 ISR이 유도되었으며, 이러한 저항성 반응이 식물 호르몬 중 에틸렌과 자스몬산에 의한 것임을 실험을 통해 증명하였다(Weller 등, 2012). P. aeruginosa 7NSK2가 생성하는 pyocyanin도 ISR을 유도하는 항생물질로 알려져 있으며, Botrytis cinerea에 대해 토마토의 저항성 반응을 유도하며 이때 pyocyanin 생성과 관련된 phzM 유전자가 중요한 역할을 하는 것으로 알려졌다(Audenaert 등, 2002).

② 생물계면활성제(biosurfactants)

미생물이 균체 외에 생성하는 화합물 중 친수성과 소수성을 모두 가지는 물질(amphiphilic compound)을 생물계면활성제(biosurfactants)라 한다(Carrillo 등, 1996). 최근 들어 식물의 ISR을 결정하는 요인 중 하나로 생물계면활성제가 주목 받고 있는데, 특히 짧은 oligopeptide에 지방산 사슬이 연결된 구조인 cyclic lipopeptide (CLP)에 대한 연구가 활발히 진행되고 있다(Raaijmakers 등, 2006). 대표적인 CLP 생성 균으로는 Bacillus속과 Pseudomonas속이 있으며, 그 중 Bacillus는 크게 surfactin, fengycin, iturin 등의 CLP를 형성하는 것으로 보고되었다(Ongena 등, 2007). Surfactin과 fengycin은 콩과 토마토에서 B. cinerea에 대한 ISR을 유도하며, 특히 토마토에서는 surfactin이 더 효과적으로 작용하였다(Ongena 등, 2007). Iturin은 fengycin과 함께 지질 이중층의 원형질막에 결합해 구멍을 내어 손상을 주며, 이를 통해 다양한 병원균에 대한 항생효과를 갖는다(Falardeau 등, 2013). Pseudomonas속은 viscosin, amphisin, tolaasin, syringomycin 등의 CLP를 형성하며, 가장 많이 연구가 되어온 viscosin의 massetolide A는 토마토에서 P. infestans에 대해 직접적인 길항작용을 할 뿐만 아니라 식물의 저항성 유도에 관여한다고 보고되었다(Tran 등, 2007). Rhamnolipids는 Pseudomonas속과 Burkholderia속이 형성하는 생물계면활성제로, 양분 흡수 및 생물 분해와 biofilm을 형성하는 중요요소로 알려져 있다(Abdel-Mawgoud 등, 2010). Rhamnolipids는 다양한 병원균 B. cinerea, H. arabidopsidis, P. syringae pv. tomato에 대해 애기장대에 저항성을 유도하며, 살리실산이 저항성 유도 과정에서 필요요소로 작용한다고 보고되었다(Sanchez 등, 2012). 특히 P. aeruginosa가 형성하는 Rhamnolipids는 포도에 B. cinerea에 대한 저항성을 유도하는데 활성산소종을 형성하거나 저항성 유전자 발현을 유도하여 병을 감소시킨다고 보고되었다(Varnier 등, 2009).

③ 휘발성 물질(volatile organic compounds, VOCs)

다양한 미생물에 의해 분비되는 VOCs는 식물의 발달과 식물-미생물 및 식물-곤충의 상호작용에도 중요한 역할을 수행한다(Tarkka와 Piechulla, 2007). VOC는 병원균의 생장을 직접적으로 억제시키며 식물에 저항성을 유도하여 간접적으로 생물적 스트레스로부터 식물의 피해를 줄이기도 한다(Ryu 등, 2004). Ryu 등(2004)B. subtilis GB03, B. amyloliquefaciens IN937a가 분비하는 물질인 2,3-butanediol과 3-hydroxy-2-butanone에 의해 애기장대에서 세균성 무름병원균 Erwinia carotovora subsp. carotovora SCC1에 대한 저항성이 증가한다고 보고하였다. Flavobacterium johnsoniae GSE09이 분비하는 2,4-di-tert-butylphenol은 P. capsiciColletotrichum acutatum의 생장을 억제시킬 뿐만 아니라, 저항성을 유도하여 식물의 역병 및 열매의 탄저병 피해를 감소시켰다(Sang과 Kim, 2012; Sang 등, 2011).

④ 비타민(vitamin)

비타민은 동 · 식물의 물질대사 과정을 조절하며 관련 반응을 돕는 중요한 물질이다(Palacios 등, 2014). 식물은 대사과정에 필요한 비타민을 스스로 생합성할 수 있지만 일부 중요 비타민을 생성하지 못하기도 하는데, 이때 비타민을 생합성하는 근권미생물과의 상호작용을 통해 필요한 것을 보충 받기도 한다(Baya 등, 1981; Campbell 등, 2006; Miyamoto 등, 2002). 또한, 비타민은 다양한 병원균에 대한 저항성을 유도하기도 하며, 저항성 유도 기작에 직 · 간접적으로 관여하기도 한다(Boubakri 등, 2016). Azotobacter vinelandii, P. fluorescens 등의 다양한 근권미생물은 riboflavin (vitamin B2)을 생성하는데, riboflavin은 애기장대의 Peronospora parasiticaP. syringae pv. tomato DC3000, 담배의 A. alternataTobacco mosaic virus (TMV)에 대한 저항성을 유도하며, R. solani에 대한 저항성 반응에서 활성인자(activator)로써의 역할을 수행한다고 알려졌다(Dong과 Beer, 2000; Zhang 등, 2009). 또한 riboflavin은 식물의 H2O2의 축적을 유도하고 이를 통해 hypersensitive cell death를 유도하여 식물 저항성 반응에 관여하고 있는데(Azami-Sardooei 등, 2010), 최근 미생물 인자의 저항성 유도 기능을 이용한 제품화가 다양하게 진행되고 있으며 riboflavin을 함유한 바이오닥터의 경우 벼 도열병과 흰잎마름병을 억제시키는 효과가 있다고 보고되기도 하였다(Kang 등, 2016a). B. subtilis 등의 미생물이 생성하는 pyridoxine (vitamin B6)은 병원균 Cercospora nicotianae로부터 식물을 보호하며, 염 · 광산화 등 환경 스트레스에 대한 내성을 함께 유도하였다(Bilski 등, 2000; Chen과 Xiong, 2005). Ascorbic acid (vitamin C)는 식물의 광합성, 세포 생성 및 생장 촉진, 호르몬 생합성에 관여하며(Palacios 등, 2014), 애기장대에서 SAR 기작인 SA-의존적 면역반응의 유전자들의 활성과 연관이 있다고 보고되었다(Barth 등, 2004; Pastori 등, 2003).

생물 스트레스에 대한 식물 신호전달 체계 변화에 미치는 미생물 영향

앞에서 언급한 다양한 미생물 인자를 식물이 인식하게 되면, 식물은 살리실산(salicylic acid, SA), 자스몬산(jasmonic acid, JA), 에틸렌(ethylene, ET) 등의 호르몬 시스템을 조절하여 신호전달체계 변화를 유도한다(van Loon 등, 1998). 이러한 호르몬은 식물체 내에서 충분히 축적되어야만 다양한 병원균에 대한 저항성을 유도할 수 있다(Ryals 등, 1996). 식물의 대표적인 유도저항성 반응인 SAR과 ISR은 넓은 범위의 병 방제 효과로 인하여 유사한 기작을 갖는다고 생각되어 왔으나 SA의 생합성 및 pathogenesis-related (PR) 유전자의 발현양상에서 서로 간의 차이가 존재하였다(Hoffland 등, 1995). P. fluorescens WCS417r을 처리한 radish에서 유도되는 F. oxyporum에 대한 저항성에서 SAR의 특성인 PR protein의 생성이 일어나지 않았고, SA 생합성이 불가능한 애기장대 돌연변이 NahG 식물 실험을 통해 ISR은 SA와는 다른 독립적인 기작을 통해 저항성이 유도되는 것을 알 수 있었다(Hoffland 등, 1995; Pieterse 등, 1996, 2000). 근권미생물에 의한 ISR 반응과 SA 기작 간의 연관성에 대한 연구가 계속해서 진행되던 중, P. aeruginosa 7NSK2와 SA를 생성하는 돌연변이 균주 P. aeruginosa KMPCH를 통해 SA도 ISR에 관여할 수 있다는 결과가 보고되었으며(De Meyer 등, 1999), 일부 Bacillus속 세균의 경우에서도 ISR이 일어나기 위해서는 SA의 생합성을 필요로 한다는 것이 알려졌다(Barriuso 등, 2008). 따라서 근권미생물에 의한 식물의 인식과 신호전달 등의 과정은 유동적이며, 같은 ISR 반응이라도 병원균 또는 근권미생물에 따라 신호전달 방식이 다를 수 있다(Conn 등, 2008).

SA와 마찬가지로 JA와 ET 역시 식물의 ISR 신호전달에 관여하는 중요한 조절인자인데(van Loon 등, 1998), JA 및 ET 신호전달 관련 애기장대 돌연변이 연구를 통해 두 호르몬이 근권미생물에 의해 유도되는 ISR의 핵심 물질로 알려졌다(Pieterse 등, 1998). JA 신호전달 관련 돌연변이 jar1, jin1, coi1 애기장대 식물 및 ET 신호전달 관련 돌연변이인 etr1, ein2, ein3, eir1과 근권세균 P. fluorescens WCS417r, Serratia marcescens 90-166, Penicillium sp. GP16-2, Trichoderma harzianum T39의 상호작용을 통해 SA와는 독립적으로 JA와 ET가 ISR에 관여한다고 보고되었다(Pieterse 등, 2014; Pozo 등, 2008). 하지만 애기장대의 JA 신호전달 관련 돌연변이 jar1과 ET 신호전달 관련 돌연변이 etr1, SAR 기작 관련 돌연변이 npr1을 이용한 실험을 통해 PGPR P. fluorescens WCS417r이 P. syringae pv. tomato DC3000에 대하여 기존에 알려진 것처럼 JA와 ET 관련 신호전달체계를 가지면서, 반면에 SAR과 유사하게 NPR1에 의해서 조절됨으로써 NPR1이 저항성이 유도되는 신호물질에 따라 다르게 방어 반응을 조절하는 cross-talk을 하는 것으로 밝혀졌다(Pieterse 등, 1998, 2000).

Brassinosteroid (BR)는 식물의 세포 및 줄기 생장, 화분관(pollen tube) 형성 등을 통해 식물의 생육을 촉진시키면서(Azpiroz 등, 1998; Ryu 등, 2005) 병 저항성을 유도하는 호르몬으로 알려져 있다(Khripach 등, 2000). Nakashita 등(2003)은 BR 관련 기작이 담배의 TMV, P. syringae pv. tomato DC3000에 대한 저항성 반응을 유도하면서 벼의 도열병, 흰잎마름병에 대한 저항성을 유도한다고 보고하였다. B. subtilis GB03은 VOCs를 통해 애기장대에서 생육촉진 및 ISR을 유도하는데, BR을 인식하지 못하는 돌연변이 식물 cbb1에서는 그 효과가 일부 감소한다는 것을 관찰하였다(Ryu 등, 2005). 이처럼 식물의 BR 인식 기작은 유용 미생물과의 상호작용에 있어서 중요한 역할을 하는데, 최근 BR의 수용체 BRI1 (BRASSINOSTEROID-INSENSITIVE 1) 기작과 관련된 BAK1 (BRI ASSOCIATED RECEPTOR KINASE 1)이 다양한 미생물들을 인식하는 MAMP 수용체와의 상호작용을 통해 초기 면역반응을 이끌어 낸다고 보고되었다(Shan 등, 2008). Azelaic acid (AZA)는 식물체 내 SA의 축적을 통해 방어 프라이밍을 유도하는 데 관련된 신호전달물질로 식물 면역반응에 있어서 중요한 역할을 하는 것으로 알려졌다(Jung 등, 2009). 다양한 식물에 병저항성을 유도하는 근권미생물 Azospirillum brasilense (Bashan과 De-Bashan, 2002)을 처리한 애기장대에서 AZA 기작과 관련된 AZELAIC ACID INDUCED 1 (AZA1)의 발현이 증가한 것이 보고되었으나 스트레스 저항성 유도와 관련된 기작에 대한 연구는 더 필요하다(Spaepen 등, 2014).

식물의 환경 스트레스에 대한 미생물의 역할

식물 환경 스트레스 내성 유도

PGPR 등의 근권미생물은 IST를 유도하여 다양한 환경 스트레스에 대해 식물을 보호한다(Yang 등, 2009). PGPR 등의 근권미생물은 염류 스트레스에 의한 식물체 내 높은 에틸렌 함량을 조절하여 정상적인 생육을 가능하도록 해주며(Glick 등, 2007a, 2007b), 토양 나트륨, 칼슘 이온 등의 흡수 패턴을 변화시키거나(Nadeem 등, 2007) 물 흡수율을 증가시켜 염류 스트레스로부터 식물에 내성을 유도하기도 한다(Yue 등, 2007). Bensalim 등(1998)은 고온 조건에서 근권세균(rhizobacteria)을 처리한 감자에서 고온에 대한 내성이 유도되는 것을 확인하였다. 또한 유용한 근권미생물에 의해 저온조건에서 식물의 생중량(biomass) 감소율이나 전해질 누출량이 감소하여, 식물이 스트레스 조건에 빠르게 적응할 수 있도록 도와주는 것으로 생각된다(Ait Barka 등, 2006). 그밖에 건조 스트레스나 토양의 영양소 부족, 중금속 독성 등의 환경 스트레스 조건에서도 식물에 내성을 유도하여 식물을 보호하는 것으로 생각된다(Carrillo-Castañda 등, 2003, 2005; Egamberdiyeva, 2007).

환경 스트레스 감소에 영향을 주는 미생물 결정 인자

① 식물호르몬(phytohormone)

PGPR 등의 근권미생물은 식물호르몬을 생성 또는 분해하며 다양한 기작을 통해 식물 생육에 영향을 주고 있다(Dodd 등, 2010). 식물호르몬은 환경 스트레스와 같은 부적절한 조건에서 식물이 견딜 수 있도록 도와주는 역할을 하며(Fahad 등, 2015), 미생물이 식물호르몬을 직접 생성하여 기주식물 세포의 생장과 분해를 도와 환경 스트레스 조건에서도 식물 생육에 도움을 주는 것으로 보고되었다(Dodd 등, 2010). 식물호르몬으로 알려진 지베렐린(gibberellin, GA)은 일반적인 조건에서뿐만 아니라 염 및 건조 등의 다양한 환경 스트레스 조건에서 식물의 생육을 촉진하는 데 관여하였고(Hamayun 등, 2010; Radhakrishnan 등, 2013), GA를 생성하는 PGPR인 P. putida H-2-3은 건조한 환경에서도 콩의 생육을 증가시켰다(Kang 등, 2014). 앱시스산(abscisic acid, ABA)은 건조조건에서 수분 손실에 대해 식물의 기공 및 삼투압을 조절하는 데 관여하는 대표적인 호르몬으로(Kaushal과 Wani, 2016; Keskin 등, 2010), 실제 ABA를 형성하는 근권세균 Azospirillum은 애기장대, 옥수수, 밀 등에서 토양 건조 스트레스에 대한 내성 증가를 유도하였다(Cohen 등, 2009; Dodd 등, 2010). 이 외에도 사이토카인(cytokine)을 생성하는 B. subtilis는 측백나무(Platycladus orientalis) 내 ABA의 양을 조절하여 환경 스트레스에 대한 내성을 유도하였으며(Liu 등, 2013), 식물의 생육 및 뿌리생장을 유도하는 indole-3-acetic acid (IAA)도 환경적 스트레스 내성을 유도하는 미생물 결정 인자 중 하나로 알려졌다(Vurukonda 등, 2016). Yuwono 등(2005)의 연구에 따르면, 건조 조건에서 IAA를 처리한 벼의 뿌리 생장률이 증가하였으며, IAA를 생성하는 Azospirillum을 처리한 식물에서도 유사한 현상이 관찰되었다(Dimkpa 등, 2009). 하지만 고농도의 IAA는 ET의 생합성을 촉진시켜 식물 뿌리 생장을 억제시키는 작용을 하기 때문에, PGPR에 의한 IAA의 생성 농도도 식물의 생육 및 스트레스 내성을 유도하는 데 중요한 요인이 될 수 있다(Dimkpa 등, 2009; Eliasson 등, 1989).

② 삼투물질(osmolyte)

수분 손실을 야기하는 조건에서, 미생물의 삼투물질 분비 및 축적은 식물의 수분 스트레스 내성 유도에 중요한 기능을 한다(Vurukonda 등, 2016). 대표적인 삼투물질로는 proline, sugars, polyamines, betaines, quaternary ammonium compounds, polyhydric alcohols와 기타 amino acid, 그리고 dehydrin과 같은 수분 스트레스 단백질이 있다(Yancey 등, 1982). 삼투물질은 미생물이 분비하거나 식물체 내에서 합성되어 환경 스트레스에 대해 시너지로 내성을 유도하거나 식물의 생육을 촉진시키기도 한다(Paul 등, 2008). PGPR인 Bacillus polymyxa는 proline을 분비하여 토마토의 건조 스트레스 내성을 유도하며(Shintu와 Jayaram, 2015), Burkholderia phytofirmans PsJN에 의해 분비된 proline은 저온 스트레스에 대한 내성도 유도한다고 알려졌다(Ait Barka 등, 2006). 그 외에 choline, trehalose 등의 물질은 삼투억제제(osmoprotectants)의 기능을 수행하며, 다양한 PGPR들이 삼투억제제를 생성 및 분비하여 염 또는 건조스트레스에 대해 식물에 내성을 유도하였다 (Figueiredo 등, 2008; Qurashi와 Sabri, 2011; Suárez 등, 2008).

③ Exopolysaccharide

세균이 외부로 분비하는 exopolysaccharide (EPS)는 주로 탄소화물 중합체(carbohydrate polymers)로 구성되어 있으며, 세균이 흙에 부착하거나 응집할 때 중요한 역할을 하는 것으로 알려졌다(Flemming과 Wingender, 2001). EPS는 생물적 스트레스로부터 식물을 보호하며(Haggag, 2007; Sankari 등, 2011), 환경 스트레스 내성 증가에도 도움을 주는 것으로 보고되었다(Dimkpa 등, 2009). PGPR을 비롯한 세균은 스트레스 조건에서 EPS 생성량이 증가하는데, 이를 통해 EPS 생산 반응이 세균이 스트레스에 대응하기 위한 과정이라는 것을 알 수 있었다(Roberson과 Firestone, 1992). 분비된 EPS 물질은 캡슐형태로 비교적 끈적한 성질을 가지며, 식물의 뿌리 주변 토양을 입단화(aggregation)시키는 데 관여하여 토양 구조를 변형시키고, 변형된 입단구조는 높은 수분포텐셜을 유지하거나 건조되는 속도를 낮춰 토양 내 미생물과 식물을 건조 스트레스로부터 보호하는 데 도움을 준다(Choudhary 등, 2016). Alami 등(2000)의 연구에 따르면, 건조 스트레스에서 해바라기의 뿌리 조직에 부착되어 있는 토양의 비율을 측정한 결과 EPS를 생성하는 근권세균인 YAS34를 접종한 식물에서 뿌리에 붙어 있는 토양의 양이 증가한 것을 알 수 있었다. 또한 Proteus penneri, P. aeruginosa, Alcaligenes faecalis 등 EPS를 생성하는 균주를 옥수수 종자에 처리하면 식물의 뿌리나 줄기 길이, 잎의 면적 등 식물 생육을 증가시켰을 뿐만 아니라, 건조 스트레스 조건에서 상대 수분함량, 단백질 및 당 함량, 항산화효소(antioxidant enzymes)의 활성이 증가하였다(Nassem과 Bano, 2014). 염 스트레스 조건에서는 다양한 EPS 생성 균주를 접종한 밀의 뿌리에서 나트륨 이온 흡수율이 감소되었고, 이를 통해 EPS가 식물의 염 스트레스 내성에도 관여하고 있다는 것을 확인하였다(Ashraf 등, 2004).

④ 휘발성 물질(volatile organic compounds, VOC)

PGPR 등의 유용한 근권미생물이 생성하는 VOCs는 ISR을 통해 식물의 생물적 스트레스에 대한 저항성을 유도하면서 IST를 통해 환경 스트레스에 대한 내성을 유도하기도 한다(Choudhary 등, 2016). 염 스트레스 환경에서 식물 뿌리의 나트륨 이온 흡수율은 식물이 스트레스에 대한 내성을 갖는 데 중요한데, B. subtilis GB03이 분비하는 VOCs는 애기장대의 뿌리에서 흡수에 관여하는 유전자인 HIGH-AFFINITY K+ TRANSPORTER 1 (HKT1)의 발현을 조절하여 염 스트레스에 대한 IST를 유도하였다(Zhang 등, 2008). 또한 VOCs 물질 중 하나인 2R, 3R-butanediol을 분비하는 P. chlororaphis O6는 건조 스트레스 조건에서 애기장대의 기공 개폐를 조절하여 식물의 수분손실을 막도록 유도함으로써 식물의 건조내성을 증가시킨다고 보고하였다(Cho 등, 2008). 최근에는 근권미생물에 의해 유도되어 배출되는 식물의 VOC도 환경적 스트레스에 대한 내성을 유도할 수 있다는 사실이 알려졌다(Timmusk 등, 2014). B. thuringiensis AZP2를 처리한 밀은 건조 스트레스 조건에서 무처리 식물보다 생중량 및 광합성량이 증가하였고 스트레스 조건에서 5배 이상의 생존율이 유지되었는데 이때 식물 잎에서 배출되는 benzaldehyde, β-pinene, geranyl acetone의 VOC양이 증가한 것과 관련이 있을 것으로 알려졌다(Timmusk 등, 2014).

환경 스트레스 관련 식물 신호전달 체계에 미치는 미생물 영향

스트레스와 관련된 식물호르몬의 체내 축적은 식물 생육에 영향을 줄 뿐만 아니라 다양한 환경 스트레스 조건에 대한 식물의 내성을 증가시킨다(Maksimov 등, 2015). ABA를 형성하는 A. lipoferum USA 59b는 식물 내 ABA 함량을 높여 건조 스트레스 조건에서도 식물의 생육을 촉진시킴으로써 내성을 증강시켰고, ABA 생합성 돌연변이 토마토에 근권세균 B. megaterium을 처리할 경우, 돌연변이 토마토 유묘의 ABA 함량이 최적으로 유지되면서 다양한 환경 스트레스에 대한 내성이 유도되었다(Cohen 등, 2009; Porcel 등, 2014). Timmusk와 Wagner (1999)의 연구에 따르면, Paenibacillus polymyxa를 처리한 애기장대에서 ABA-의존적 기작에 관여하는 유전자 EARLY RESPONSE TO THE DEHYDRATION 15 (ERD15)의 발현이 증가하여 건조 스트레스 내성이 강하게 유도되는 것이 관찰되었다. 식물 열매의 숙성과 밀접한 관련이 있는 호르몬인 ET는 식물의 활성을 조절하고 있으며, 생물 · 환경 스트레스에 반응하여 체내에서 생합성이 조절되기도 한다(Hardoim 등, 2008). ET는 식물의 종이나 세포 타입에 따라 생육을 촉진시키기도 하고 억제시키기도 하는데, 특히 스트레스 조건에서는 ET가 식물체 내에서 항상성(homeostasis)을 조절함으로써 식물의 줄기 및 뿌리 생장을 감소시켰다(Dodd와 Perez-Alfocea, 2012; Pierik 등, 2007). PGPR을 비롯한 다양한 근권미생물은 ET의 전구체인 1-aminocyclopropane-1-carboxylate (ACC)를 분해하는 효소인 ACC deaminase를 생성하는데, 세균에 의해 생성되는 ACC deaminase는 식물체 내 ET에 의한 영향을 감소시켜 환경 스트레스를 완화시키는 데 중요한 역할을 수행한다(Glick, 2006). ACC deaminase를 생성하는 Pseudomonas가 염류 조건 또는 건조조건에서 작물 열매의 성숙을 촉진시키며 수확량을 증가시킨다고 보고되었으며, Bacillus, Variovorax 등 ACC deaminase를 생성하는 다양한 PGPR이 식물의 환경 스트레스 완화 및 내성 유도와 연관이 있었다(Ahmad 등, 2011; Arshad 등, 2008; Saleem 등, 2007).

식물의 생물 · 환경 복합 스트레스 경감에 미생물 역할

생물 또는 환경적 스트레스에 대한 내성을 유도하는 근권미생물에 대한 연구는 꾸준히 진행되어 왔으며, Timmusk와 Wagner (1999)는 복합 스트레스 경감에 미생물을 적용할 가능성을 열어주었다. P. polymyxa는 애기장대에서 생물 스트레스(Erwinia carotocora에 의한 무름병)를 감소시키며, 미생물을 처리한 애기장대에서 건조 스트레스 내성에 관여하는 유전자 EARLY RESPONSE TO THE DEHYDRATION 15 (ERD15)(Kiyosue 등, 1994)의 발현량이 무처리 식물에 비해 약 50배 이상 증가하여 복합 스트레스 저항성에 관여한다고 생각되었다(Timmusk와 Wagner 등, 1999). 이후 ERD15는 염류 등 다양한 환경 스트레스에 대한 내성을 유도한다고 추가적으로 알려졌으며, ERD15 과발현 돌연변이 실험을 통해 해당 유전자가 SA-의존적 면역반응에 관여함으로써 E. carotovora에 대한 저항성을 유도한다는 것이 밝혀졌다(Kariola 등, 2006; Ton 등, 2009). PGPR 등의 근권미생물이 생성하는 EPS는 건조 또는 염 스트레스에 대한 내성을 유도하거나 스트레스를 완화시키기도 하지만(Ashraf 등, 2004; Choudhary 등, 2016), 진균 병원균인 Aspergillus niger의 생장을 직접적으로 저해하거나 식물에 저항성을 유도하기도 하였다(Haggag, 2007). PGPR이 생성하는 VOCs (Ryu 등, 2004; Zhang 등, 2008), ACC deaminase (Gontia-Mishra 등, 2014), biofilm (Bogino 등, 2013) 등 다양한 물질들이 두 스트레스에 대한 내성을 유도한다고 보고되었다. P. aeruginosa PW09를 종자 및 유묘에 처리한 오이는 S. rolfsii에 대한 저항성이 증가하였으며 동시에 염 스트레스 조건(150 mM)에서도 무처리 식물에 비해 높은 내성을 보였다(Pandey 등, 2012). Lucas 등(2014)의 연구에 따르면, Chryseobacterium sp. BaC1-13, Pseudomonas sp. BaC1-21, BaC1-38의 근권미생물을 이용해 ISR을 유도한 벼에서 X. campestris에 대한 저항성과 염 스트레스에 대한 내성이 함께 유도되었으며, 식물 내 다양한 2차 대사산물의 생성이 증가하였다. 단일 미생물뿐만 아니라 근권미생물 군집도 식물-토양-미생물 상호작용을 통하여 효과적으로 식물에 영향력을 줄 수 있는 요인 중 하나이다(Chaparro 등, 2012). 근권미생물은 식물의 뿌리에서 나오는 exudate의 조성에 영향을 주며, 이는 다시 근권미생물이나 토양 조성에 영향을 줄 수 있는데(Badri 등, 2009), 식물 근권미생물 군집은 식물의 영양분 흡수를 용이하게 해줄 뿐만 아니라 생물 · 환경 스트레스로부터 식물을 보호하기도 한다(Mendes 등, 2013). 이처럼 근권미생물 군집은 다양한 식물 스트레스에 대해 영향을 주며, 이에 대한 연구는 앞으로 미생물을 농업생태계에 안정적으로 적용시키는 데 중요한 역할을 할 것이다(Rodriguez 등, 2008).

결언

실제 농가에서는 급격하게 변화하는 기후와 다양한 재배환경의 변화로 생물적 스트레스와 환경 스트레스가 동시에 일어나 식물을 생산하는 데 어려움을 겪고 있다. 이와 같은 복합 스트레스는 각각의 단일 조건보다 더 심각한 피해를 불러올 수 있으며, 단일 스트레스에서는 유도되지 않던 다양한 전사 관련 유전적 반응들이 새롭게 일어날 수 있다. 이렇게 다양하게 변화하는 환경에서 친환경적으로 지속가능한 농업을 유지하기 위하여 근권미생물을 이용한 방안이 대두되고 있으며, 앞으로는 근권미생물을 이용한 단일 스트레스 저항성 또는 내성 유도뿐만 아니라, 생물 · 환경 스트레스가 동시에 일어나는 복합 스트레스에 대한 연구도 필요할 것이다. 생물 · 환경의 복합 스트레스에 대한 내성을 동시에 유도하는 근권미생물의 효과와 관련 기작에 대한 연구를 이어간다면, 실제로 기후변화에 따른 농가의 피해를 감소시킬 수 있는 효과적인 대응 방안으로 이어질 수 있을 것이라 생각된다.

요약

식물은 재배기간 동안 세균, 진균, 바이러스 등의 생물 스트레스뿐만 아니라 고온, 염, 건조 등 다양한 환경 스트레스에도 노출되어 왔다. 최근에는 기후 이상현상으로 인하여 환경 스트레스의 빈도 및 강도가 불규칙적으로 증가하고 있으며 이로 인해 병원균의 생장과 영향도 변화하여 생물과 환경의 복합 스트레스가 식물 재배에 큰 영향을 주고 있다. 유용미생물을 이용한 식물의 저항성 유도는 다양한 생물과 환경 스트레스로부터 식물을 보호하는 데 도움을 주며, 이러한 스트레스에 대한 피해를 감소시킬 수 있는 가능성을 열어 주었다. 본 리뷰에서는 식물의 생물과 환경 스트레스에 대한 피해를 감소시키는 데 영향을 주는 미생물의 결정인자에 대해 기술하였으며 미생물 결정인자에 의해 유도되는 식물 신호전달 체계 변화에 대해 기술하였다. 또한 복합 스트레스 경감을 위한 미생물의 역할과 연구 방향에 대해 기술하였다. 이 리뷰를 통해 변화하는 환경에 대비하기 위해서 다양한 방안을 마련하고 있는 농민들에게 도움이 되기를 바라며, 실제 유용미생물 연구가 식물 재배 중 발생할 수 있는 다양한 스트레스에 따른 농가 피해를 감소시킬 효과적 대응 방안으로 이어지길 바란다.

A scheme for the effects of plant growth-promoting bacteria (PGPB) on biotic and abiotic stress in plants. A multiple stresses (bioticabiotic) can cause more serious damage to plants than each stress condition; PGPB can induce systemic resistance/tolerance or directly alleviate the stresses by production of diverse bacterial determinants. PGPB can intervene plant signal transduction including salicylic acid (SA), jasmonic acid (JA), ethylene (ET), absicisic acid (ABA), brassinosteroid (BR), or azelaic acid (AZA), and this process could help mitigate multiple stresses.
