Abstract

국내에서 재배되는 사과 ‘홍로’에 발생하는 Apple scar skin viroid병의 유전자 계통분석을 하기 위하여 바이로이드 증상이 나타나는 8개 지역(봉화, 청송, 당진, 김천, 무주, 문경, 수원, 영월)의 농가에서 수집한 시료를 RT-PCR 방법을 이용하여 바이로이드 검정을 실시하였다. 검출된 바이로이드의 클로닝과 유전자 염기서열 분석을 통해 8종의 분리주들을 확인하였고 한국, 인도, 중국, 일본 및 그리스에서 보고된 21종의 분리주와 염기서열을 비교하였다. 8개의 분리주들의 핵산 서열은 기존에 보고된 분리주들과 92.2-99.7%의 상동성을 나타냈다. 계통분석을 통해 본 연구에서 보고한 7종의 분리주들은 기존의 것들과 독립된 그룹에 속하는 것으로 나타났다.

Trans Abstract

To identify genome sequences of Apple scar skin viroid (ASSVd) isolates in Korea, the field survey was performed from ‘Hongro’ apple orchards located in eight sites in South Korea (Bongwha, Cheongsong, Dangjin, Gimchoen, Muju, Mungyeong, Suwon, and Yeongwol). ASSVd was detected by RT-PCR and PCR fragments were cloned into cloning vector. Full-length viral genomes of eight ASSVd isolates were sequenced and compared with 21 isolates reported previously from Korea, India, China, Japan and Greece. Eight isolates in this study showed 92.2-99.7% nucleotide sequence identities with those reported previously. Phylogenetic analysis showed that seven isolates reported in this study belong to the same group distinct from other groups.

서론

사과(Malus domestica)는 장미과 사과나무속에 속하는 과수 작물로 국내 총 재배면적은 30,000 ha이며 연간 475,000톤이 생산되고 있는 대표적인 소득작물이다(Ministry of Agriculture, Food and Rural Affairs, 2015). 국내 사과에서 발생하는 주요 바이러스 및 바이로이드병으로는 Apple mosaic virus, Apple chlorotic leaf spot virus, Apple stem pitting virus, Apple stem grooving virus, Apple dimple fruit viroid, Apple scar skin viroid(ASSVd) 등이 있으며, 이러한 병에 감염될 경우 수량 감소와 품질 저하의 문제가 나타나는 것으로 알려져 있다(Lee, 2013).

바이로이드는 약 250-400 nucleotides(nt)의 환형 단일가닥 RNA로 구성되어 있으며, 일반적인 바이러스와 달리 핵산을 싸고 있는 외피 단백질이 없고 어떠한 단백질도 코딩하고 있지 않아 오로지 기주 내 여러 인자들의 도움을 받아서 복제가 이루어 진다(Diener, 1987; Di Serio 등, 1996; Hashimoto와 Koganezawa, 1987). ASSVd는 국내 재배 사과에 가장 심각한 피해를 끼치는 대표적인 사과 바이로이드로서, 기주에 단독 혹은 복합 감염될 경우 잎과 과실에 모자이크, 기형 등 병징을 나타내며, 결국 생산량이 감소하게 되는 것으로 알려져 있다(Hashimoto와 Koganezawa, 1987; Zhu 등, 1995). ASSVd는 중국에서 1935년 처음 발견이 되었으며(Ohtsuka, 1938), 일본, 미국, 인도, 유럽 등지에서도 보고된 바 있다(Cambell과 Sparks, 1976; Diener, 1987; Millikan과 Martin, 1956; Yamaguchi와 Yanase, 1976). 국내에서는 2001년 사과 재배농가에서 일본 도입종인 ‘미끼라이프’ 품종에서 ASSVd의 발생이 최초 보고되어 ASSVd의 일부 구조에 대한 연구(Lee 등, 2001)가 이루어졌고, 군위, 상주, 무주 등 사과 주산지에서의 ASSVd 발생 양상에 대한 연구가 수행되었다(Kwon 등, 2002). 하지만, 국내 분리주들을 대상으로 지역 별로 전체 핵산 서열을 분석하고, 지역에 따른 분리주 서열들간의 계통분석을 진행하거나 해외에서 보고된 분리주 서열들과 비교를 진행한 연구는 진행된 바 없었다. 본 연구는 2014년부터 2015년까지 국내 사과 주산지 8개 지역을 중심으로 수집한 ASSVd의 분리주들에 대한 전체 핵산서열들을 보고하고, 이에 대한 계통 분석을 통해 국내 분리주 및 해외에서 기 보고된 분리주들의 서열들 간의 차이를 확인하여 ASSVd 연구의 기초자료로 활용하고자 연구를 수행하였다.

ASSVd 검정

2014년 4월부터 2015년 3월까지 사과 주산단지인 수원, 봉화, 청송, 당진, 김천, 무주, 문경, 영월 8개 지역 32 곳의 재배농가에서 ‘홍로’ 품종의 사과나무 20주를 무작위로 선발하고, 총 640개 사과나무에서 1개 사과나무당 4-5점의 잎 샘플을 묶어서 채취하여 각각의 나무에 대한 바이로이드 검정에 사용하였다(Table 1). Total RNA는 CTAB 방법을 이용하여 추출 하였으며(Gambino 등, 2008), RT-PCR은 기존에 보고된 ASSVd 의 특이적 프라이머인 AS3(5’-CCCGGTAAACACCGTGCGGT-3’) 와 AS4(5’-ACCGCGAAACACCTATTG TG-3’)를 이용하여 수행하 였다(Lee 등, 2001). ASSVd가 검출된 이병주 과실에서 8-9월경에 얼룩반점 증상이 관찰되었으며(Fig. 1), 과실의 크기와 수확 기가 늦어지는 증상을 확인할 수 있었다(자료생략).

ASSVd 유전자 분석 및 계통분석

ASSVd가 검출된 지역 에서 무작위로 선발한 이병시료의 정확한 동정을 위하여 지역당 5개씩 RT-PCR 산물을 pGEM-T easy vector (Promega, USA) 클로닝하여 염기서열을 분석한 결과 331 nt 길이의 ASSVd 전체 염기서열을 확인하였다(Fig. 2). 지역별로 공통된 서열을 보인 분리주를 하나씩 선발하였으며, 이렇게 확인된 8종의 분리 주에 대한 서열을 NCBI GenBank에 등록하였다. 이후 이들 분리 주들의 핵산서열과 NCBI GenBank에 등록된 기존 분리주 서열들과 핵산 서열의 유사성을 MultAlin 프로그램(http://multalin. toulouse.inra.fr/multalin/) 와 MEGA6(Tamura 등, 2013)을 이용하여 비교해 본 결과, 92.2-99.7%의 핵산 서열 유사성을 보였으며, 가장 서열이 상이한 영월 분리주의 경우 다른 7개 분리주와 96.9%(김천)에서 97.5%(봉화, 문경) 사이의 유사성을 보였다 (Fig. 3, 4). 또한 해외에서 보고되었던 20종의 분리주와 기존 국내에서 보고되었던 1종의 분리주(ASSVd-K, AF421195)와 함께 총 29종 분리주 핵산 서열 유사성을 분석해 본 결과, 기존에 보고되었던 21종의 분리주와 새롭게 확인된 8종의 분리주들 간에는 92.2-99.7%의 유사성을 보이는 것으로 확인되었으며, 특히 인도(AM993159), 중국(EU031496) 분리주들과 영월 분리주는 92.2%의 가장 낮은 유사성을 보였다(Fig. 4). 총 29종의 분리주는 5개의 그룹으로 분류되는 것을 확인할 수 있었으며, 이를 바탕으로 ASSVd에 대한 새로운 분류를 제안하였다. 본 연구를 통해 새롭게 확인된 8종의 국내 ASSVd 분리주들 중 7종의 경우 기존의 분리주와 다른 C 그룹에 속하였고, 영월에서 확인된 분리주(KP765435)는 중국과 인도에서 보고된 살구와 사과의 ASSVd 분리주와 같은 E 그룹에 속하는 것으로 나타났다(Fig. 5). 계통분석 상으로는 지리적 분포에 따른 특별한 차이는 보이지 않았다. 또한, 기존에 국내에서 보고된 서열과 비교하였을 때 병원성 영역(pathogenic domain)과 중심 영역(central domain) 을 제외한 구역에서의 핵산 서열 차이를 확인하였으며, 이는 자 체적인 유전자 변이가 발생하였거나 새로운 분리주들이 유입 되었을 가능성이 있지만 이에 대해서는 추가적인 분석이 필요 할 것이다. 앞으로 국내 사과 이병주에서 검출된 바이로이드 분리주들이 사과 품질과 수량에 어떤 영향을 미치는지에 대해서도 추가적인 연구가 필요할 것이다.

본 연구는 2014년에서 2015년까지의 국내 사과 바이로이드 ASSVd 분리주의 계통분석에 대한 보고로서 바이로이드 검정기술과 이들 바이로이드 분리주들의 유전자 분석자료는 사과 등 과수에서 발생하는 바이로이드병을 지속적으로 모니터링하고 그 피해를 구명하는데 활용될 수 있을 것으로 판단된다.
