Abstract

요약

2014년 전국 무 포장의 바이러스병 발생양상과 분포조사를 실시하였다. 바이러스 병징을 나타내는 108개 시료 중 47개 시료는 TuMV가 진단되었으며 RaMV는 3개, CMV는 2개 시료에서 각각 진단되었다. TuMV와 RaMV, RaMV와 CMV의 복합감염은 나타나지 않았으나 TuMV와 CMV의 복합감염이 2개 시료에서 진단되었다. N. benthamiana에 대한 TuMV 병원성 테스트 결과, 병징의 세기에 따라 세 개의 분리주를 나누었으며 외피단백질의 아미노산 서열 분석을 통하여 약한 병징을 나타내는 분리주 R007, 강한 병징을 나타내는 분리주 R041, R065를 선발하여 계통수 분석에 이용하였다. TuMV 계통수분석과 BLAST 검색결과 TuMV 분리주 3개와 기 보고된 분리주들 간의 외피단백질 아미노산 서열은 약 98-99%의 상동성을 나타내었으며, RaMV의 외피단백질의 경우 99%의 상동성을 나타내었고, CMV의 외피 단백질은 국내 기 보고된 분리주(GenBank : GU327363)와 동일하였다. TuMV 계통수에서 R007과 R041, R065는 서로 다른 그룹에 속하였으며 그룹간의 차이는 바이러스의 기주 선호도와 관련이 있을 것이라 예상하며 추후 TuMV의 병원성 결정인자 연구에 도움이 될 것으로 사료된다.

Trans Abstract

A 2014 nationwide survey in radish fields investigated the distribution of common viruses and possible emerging viruses. Radish leaves with virus-like symptoms were collected and 108 samples assayed by RT-PCR using specific primers for Radish mosaic virus (RaMV), Cucumber mosaic virus (CMV), and Turnip mosaic virus (TuMV); 47 samples were TuMV positive, and RaMV and CMV were detected in 3 and 2 samples, respectively. No samples showed double infection of TuMV/RaMV, or RaMV/CMV, but two double infections of TuMV/CMV were detected. TuMV isolates were sorted by symptom severity, and three isolates (R007-mild; R041 and R065-severe) selected for BLAST and phylogenetic analysis, which indicated that the coat protein (CP) of these isolates (R007, R041, and R065) have approx. 98-99% homology to a previously reported TuMV isolate. RaMV CP showed approx. 99% homology to a previously reported isolate, and the CMV CP is identical to a previously reported Korean isolate (GenBank : GU327368). Three isolates of TuMV showing different pathogenicity (degree of symptom severity) will be valuable to study determinants of pathogenicity.

본문

무(Raphanus sativus L.)는 아시아 및 지중해연안이 원산지인 배추과(Cruiferae)에 속하는 작물로 우리나라의 5대 채소 중 하나이며(Lee 등 2008), 우리나라의 전체 채소 생산량 중 10-15%, 정도를 차지하고 있는 중요한 작물 중 하나이다(Ku 등, 2006). 무는 필수 무기질인 비타민 A, C와 칼슘이 풍부하여 영양학적으로 가치가 높으며 기능성 유기유황화합물인 glutathione, isothiocyanates 등의 phytochemical과 섬유소를 다량 함유하여 항암효과 및 항산화 효과를 갖는 기능성식품이다(Cheng 등, 2012). 무에 발생하는 바이러스는 Turnip mosaic virus(TuMV) 등 7종이 알려져 있으나 국내에서는 TuMV, Cucumber mosaic virus(CMV)와 Radish mosaic virus(RaMV)가 보고되어 있다(Choi 와 Choi, 1992, Kim 등, 2012). 현재까지 무 재배 농가들을 대상으로 바이러스 분포 조사가 이루어진 적은 없었으나, 1992년부터 1994년까지 강원도 일대에서 무, 배추를 대상으로 TuMV 발생 조사가 이루어졌다(Ham, 1995). 무에 감염하는 주요 바이러스 중 하나인 TuMV는 Potyviridae과(科, family) Potyvirus속(屬, genus) 바이러스로 전 세계적으로 넓게 펴져 있으며 경제적으로 많은 피해를 주는 바이러스이다. 주요 작물과 잡초들을 포함한 300종 이상의 넓은 기주 범위를 가지고 있고 89종 이상의 진딧물을 통한 비영속성 전반이 보고되고 있으나 종자전염은 되지 않는다고 알려져 있다(Raybould 등, 1999, Sánchez 등, 2003). Bromoviridae과(科, family) Cucumovirus속(屬, genus)에 속하는 CMV는 초기 바이러스 연구에서 분리, 동정된 것들 중 하나이며 1916년에 처음으로 보고되었다. CMV는 1,000종 이상의 식물을 감염시키며 최소 75종 이상의 진딧물이 비영속성으로 전파하며 종자전염 또한 보고되어 있다(Roossinck, 2001). RaMV는 Comoviridae과(科, family) Comovirus속(屬, genus) 으로 대부분의 배추과 식물에서 매개충을 통해 전염이 되며(Komatsu 등, 2007) 종자전염은 보고되어 있지 않다(Tochihara 등, 1968). 본 연구자들은 무 재배 농가에 대해 효율적인 무 바이러스 관리방안을 수립하기 위하여 TuMV, CMV와 RaMV 각각의 바이러스 분포 양상을 조사하였다. 2014년도에 도서지역을 제외한 국내 전역의 무 재배 농가에서 시료를 채집하고 바이러스 감염 실태를 조사하였다. 본 조사 결과를 바탕으로 무 재배지역의 바이러스 병 분포현황을 확인할 수 있었으며, 이에 바이러스 병 예방 및 방제 대책 수립을 위한 기초자료를 마련하였다

바이러스병 분포

바이러스가 감염된 무를 채집하기 위하여 2014년 7월부터 11월까지 전국적인 조사를 실시하였다(Fig. 1). 채집대상 지역은 경기도 이천시, 파주시, 수원시, 강원도 정선군, 양구군, 충청남도 아산시, 공주시, 당진군, 천안시, 충청북도 충주시, 보은군, 제천시, 경상남도 함양군, 경상북도 상주시, 문경시, 영덕군, 청송군, 구미시, 전라남도 광주광역시, 전라북도 무주시에서 채집하였다. 무 생산 농가 중, 주로 소규모로 무를 재배하는 포장을 대상으로 채집을 실시하였다. 울릉도나 제주도와 같은 도서 지역은 조사에서 제외하였다. 무의 바이러스 병징에는 모자이크(mosaic), 위축(dwarf), 괴사(necrosis), 얼룩(mottle), 황화(yellowing) 등이 있으며, 이러한 바이러스 병징이 뚜렷이 나타나는 무의 잎을 채집하였다. TuMV는 전형적으로 위축과 모자이크 병징을 나타내며(Fig. 2A), RaMV는 약한 황화현상과 모자이크 병징을 나타낸다(Fig. 2B). TuMV와 CMV의 복합감염은 TuMV 단독 감염시의 병징과 유사했다(Fig. 2C).

채집된 시료는 Lim 등(2005)에 제시된 방법으로 total RNA를 추출하고 TOYOBO ReverTra Ace-α-를 이용하여 cDNA를 합성하였다. 진단용 PCR 프라이머는 미국 국립생물정보센터 (National Center for Biotechnology Information)의 유전자염기서열을 참고하여 제작하였다(Table 1). PCR은 95°C 30초, 58°C 30초, 72°C 1분으로 35 cycle을 실시하여 바이러스를 진단하였다.

총 108점의 샘플 중, TuMV는 47점, RaMV는 3점, CMV은 2점, TuMV와 CMV의 복합감염은 2점으로 진단되었다(Table 2). TuMV는 8개 도 중에서 전라남도를 제외한 모든 지역에서 발생하였으며, RaMV는 전라남도에서만 발생하였고, CMV는 강원도와 충청북도에서만 발생하였다(Table 3). 전라남도에서는 단 4점의 시료만이 채집되었기 때문에 TuMV가 발생하지 않았다고 단정지을 수는 없을 것이다. TuMV는 4개월 동안의 조사기간에 전국적으로 발생됨을 확인하였다. 또한 바이러스 병징을 보였지만 위의 3종의 바이러스가 검출되진 않은 57점의 시료는 여타 바이러스의 감염 가능성이 존재하므로, 추가로 진단할 예정이다(Fig. 2D-G).

TuMV 병원성 분석 및 분리주 선정

본 연구를 위해 채집된 108개의 시료 중에서 TuMV가 진단된 47개 시료를 이용하여 모델식물(Nicotiana benthamiana)을 통해 병원성 분석 및 분리주 선정을 실시하였다. 채집된 시료의 잎을 Phosphatebuffered saline(PBS) buffer와 섞어 N. benthamiana에 2반복으로 즙액 접종을 실시하였으며, 이로 인해 나타나는 병징의 차이를 바탕으로 R007, R041, R065 3종의 분리주를 선정하였다(Fig. 3). 접종 약 3일 후, N. benthamiana에 병징이 나타나며 건전주에 비교하여 R007은 신초부위에 잎말림이 심해지며 높이 자라지 못하는 위축현상이 나타났다. R041과 R065 또한 감염 초기에는 신초에 잎말림 현상이 나타나지만 R041은 점차 엽병이 얇아지며 신초를 제외한 부분 모두가 괴사하였고, R065는 신초를 포함하여 모든 부분이 괴사하였다. N. benthamiana에 세 개의 분리주를 각각 접종하였을 때, 모든 분리주에서 병징이 심하게 나타나지만 R041, R065에 비해 R007의 병징이 상대적으로 약한 것을 관찰할 수 있었다. 이러한 병징 차이에 대한 원인을 규명하기 위해서, 세가지 분리주에 대한 외피 단백질 아미노산 서열차이를 확인하였다(Fig. 4). 아미노산 서열분석 결과, 세 가지 분리주의 아미노산의 상동성은 98.84%으로 확인되었다. 이 결과는 추후 병원성의 차이를 규명하는 연구에 이용될 수 있을 것으로 사료된다.

TuMV와 RaMV의 계통수 분석

본 연구에서 조사한 TuMV, RaMV, CMV와 기존 보고된 국내·외 분리주 간의 유전적 상관관계를 조사하기 위하여 미국 국립생물정보센터의 바이러스 외피단백질 아미노산서열을 바탕으로 각각의 계통수 분석을 실시하였다. TuMV 계통수(Fig. 5)는 크게 Group 1과 Group 2로 나뉘며 TuMV와 같은 Potyvirus속의 Potato virus Y의 외피단백질 아미노산서열을 outgroup으로 사용하였다. Group 1은 한국 등 7개 국가의 배추속(Brassica spp.) 포함 7속의 식물에서 확인 된 30개 분리주를 포함하며 Group 2는 한국 등 4개 국가의무속(Raphanus spp.) 포함 4속의 식물에서 확인된 15개 분리주를 포함한다. 계통수 분석에 사용한 전체 TuMV 분리주의 외피단백질 아미노산 서열은 평균 98.78%의 상동성을 나타내었으며 Group 1과 Group 2의 평균 상동성은 각각 99.21%, 99.28%로써 전체적으로 상당히 보존적인 서열을 공유하고 있었다. Group 1과 2를 구분 짓는 뚜렷한 지역적, 기주별 특이점은 보이지 않았다. 그러나 기주의 분포 양상을 보았을 때, Group 1 내에서는 배추속을 기주로 하는 분리주가 총 18주로 우점하고 있으며 Group 2에서는 무속을 기주로 하는 분리주가 7주, 배추속은 5주였다. 이러한 사실을 고려하였을 때, TuMV 계통수의 그룹 분리는 바이러스의 기주 선호도와 관련이 있을 것으로 사료된다. 약한 병징을 나타내는 R007은 Group 1에 속하며 AKH937J와의 상동성은 98.95%로써 가장 근연의 관계를 보였다. 반면 강한 병징을 나타내는 R041과 R065는 Group 2에 속하며 가장 가까운 분리주인 TANX2, BCNJ-1과 각각 100%, 98.61% 의 상동성을 나타내었다. 따라서 TuMV 분리주들 간의 병원성 차이는 기주 선호도와 관련이 있을 것으로 예상되며 이를 규명하기 위해서는 다양한 기주 식물을 이용한 병원성 테스트가 추가로 진행되어야 할 것이다. RaMV 계통수(Fig. 6)는 광주에 서 분리한 R061을 대상으로 체코 등 4개 국가의 배추속 작물과 무에서 분리한 6개 분리주의 large capsid protein(LCP) 아미노산서열을 비교하여 작성하였으며 RaMV와 같은 Comovirus속의 Cowpea mosaic virus를 outgroup으로 사용하였다. 전체 아미노산 서열의 평균 상동성은 98.86%로 나타났다. 본 연구에서 분리한 RaMV 국내주 R061은 대만 분리주인 TW와 가장 유연관계가 가까우며 상호간 아미노산서열은 99.47%의 높은 상동성을 나타냈으며 동일한 기주(R. sativus)에서 분리되었다. 국내·외로 RaMV 발생, 유전자 염기서열보고가 미비함을 고려 할때, 본 연구의 국내 RaMV LCP 아미노산 서열 보고와 그에 따른 계통수 분석이 국내 최초인 점에서 그 학술적 의미가 크다고 사료된다. CMV 외피단백질 아미노산서열 분석결과, R012는 국내 색동호박에서 분리된 CMV strain Z1(GenBank : GU327368)의 외피단백질과 동일하여 계통수 분석은 실시하지 않았다.

The tree was constructed with MEGA 6.0 program using Neighbor-Joining method with 1,000 bootstrap replicates. CP sequences of 7 additional isolates of RaMV for phylogenetic analysis were obtained from National Center for Biotechnology Information GenBank. The names at the end of nodes indicate Accession number/name of isolate/country/host. Black arrow indicates the isolate identified in this study. The numbers at the nodes are bootstrap values above 50%. The scale bar indicates the number of amino acid substitutions. Cowpea mosaic virus CP was used as the outgroup.
